Water-soluble\u00a0<a href=\"https:\/\/www.sciencedirect.com\/topics\/agricultural-and-biological-sciences\/polysaccharide\">polysaccharides<\/a>\u00a0(WSPs) in edible seaweeds are not only interesting\u00a0<a href=\"https:\/\/www.sciencedirect.com\/topics\/chemistry\/functional-foods\">functional food<\/a>\u00a0ingredients (eg gel-forming property, health benefits), but also useful for seaweed taxon differentiation. The current study aims to explore the utility of\u00a0<a href=\"https:\/\/www.sciencedirect.com\/topics\/agricultural-and-biological-sciences\/monosaccharide\">monosaccharide<\/a>\u00a0profiling of WSPs in seaweed differentiation. We developed a high-performance liquid chromatography-photodiode array detection for monosaccharide determination, and characterized monosaccharide profiles of WSPs from five edible seaweeds sold in Iceland (ie\u00a0<a href=\"https:\/\/www.sciencedirect.com\/topics\/biochemistry-genetics-and-molecular-biology\/laminaria\">kombu<\/a>\u00a0Iceland \u2013\u00a0<em>Laminaria hyperborea<\/em>, sugar kelp \u2013\u00a0<em><a href=\"https:\/\/www.sciencedirect.com\/topics\/agricultural-and-biological-sciences\/saccharina-latissima\">Saccharina latissima<\/a><\/em>, dulse \u2013\u00a0<em><a href=\"https:\/\/www.sciencedirect.com\/topics\/agricultural-and-biological-sciences\/palmaria-palmata\">Palmaria palmata<\/a><\/em>\u00a0and two types of\u00a0<a href=\"https:\/\/www.sciencedirect.com\/topics\/biochemistry-genetics-and-molecular-biology\/porphyra\">nori<\/a>\u00a0labeled as\u00a0<em>Porphyria<\/em>\u00a0sp. and\u00a0<em><a href=\"https:\/\/www.sciencedirect.com\/topics\/agricultural-and-biological-sciences\/pyropia\">Pyropia<\/a><\/em>\u00a0sp.). Monosaccharide profiling data reflected both the complexity and quantitative differences of WSPs. The seaweed dulse showed the highest concentrations of total sugar (ca. 16 mM), followed by kombu (ca. 12 mM). From monosaccharide profiling too\u00a0<a href=\"https:\/\/www.sciencedirect.com\/topics\/biochemistry-genetics-and-molecular-biology\/red-alga\">red algae<\/a>, it was found that\u00a0<a href=\"https:\/\/www.sciencedirect.com\/topics\/biochemistry-genetics-and-molecular-biology\/galactose\">galactose<\/a>\u00a0(ca. 81\u201387% of total sugars) and\u00a0<a href=\"https:\/\/www.sciencedirect.com\/topics\/agricultural-and-biological-sciences\/xylose\">xylose<\/a>\u00a0(ca. 65\u201377% of total sugars) are the dominant sugars in nori and dulse, respectively, which reflected the presence of\u00a0<a href=\"https:\/\/www.sciencedirect.com\/topics\/agricultural-and-biological-sciences\/galactan\">galactan<\/a>\u00a0and\u00a0<a href=\"https:\/\/www.sciencedirect.com\/topics\/agricultural-and-biological-sciences\/xylan\">xylan<\/a>. In\u00a0<a href=\"https:\/\/www.sciencedirect.com\/topics\/biochemistry-genetics-and-molecular-biology\/brown-alga\">brown algae<\/a>\u00a0(ie kombu and sugar kelp), glucose (ca. 64\u201383%) and\u00a0<a href=\"https:\/\/www.sciencedirect.com\/topics\/agricultural-and-biological-sciences\/fucose\">fucose<\/a>\u00a0(13\u201320%) are the main sugars, and they represent\u00a0<a href=\"https:\/\/www.sciencedirect.com\/topics\/agricultural-and-biological-sciences\/laminarin\">of the laminar<\/a>\u00a0and\u00a0<a href=\"https:\/\/www.sciencedirect.com\/topics\/agricultural-and-biological-sciences\/fucoidan\">fucoidans<\/a>. The\u00a0<a href=\"https:\/\/www.sciencedirect.com\/topics\/biochemistry-genetics-and-molecular-biology\/principal-component-analysis\">principal component analysis<\/a>\u00a0of sugar profiles showed the useful patterns for seaweed taxon differentiation. The\u00a0<a href=\"https:\/\/www.sciencedirect.com\/topics\/biochemistry-genetics-and-molecular-biology\/phylogenetic-tree\">dendrogram<\/a>resulting from hierarchical cluster analysis is congruent with phylogenetic tree, implying the chemotaxonomic value of seaweed monosaccharide profiles.
