Added Value of Ascophyllum nodosum Side Stream Utilization during Seaweed Meal Processing

Ascophyllum nodosum contains many valuable compounds, including polyphenols, peptides, and carotenoids that have been shown to exhibit biological activities. These compounds are not a priority ingredient in seaweed meal products for the current users. Hence, the aim of the study was to investigate the chemical and bioactive characteristics of A. nodosum as affected by seasonal variation and evaluate the potential benefits of alternative processing and the utilization of side streams for product development. The analysis of raw materials, press liquid, and press cake from alternative processing and the commercial seaweed meal at different harvesting periods indicated that the chemical composition is linked to the reproductive state of the algae. Phenolic content and ORAC activity increased following the seaweed’s fertile period, making alternative processing more promising in July and October compared to June. Several valuable ingredients were obtained in the press liquid, including polyphenols, which can be used in the development of new high-value bioactive products. The suggested alternative processing does not have a negative effect on the composition and quality of the current seaweed meal products. Hence, the extraction of valuable ingredients from the fresh biomass during the processing of seaweed meal could be a feasible option to increase the value and sustainability of seaweed processing.

Auðgaðir sjávarréttir / Enriched seafood

Verkefninu Auðgaðir sjávarréttir sem unnið var í samvinnu við fyrirtækið Grím kokk í Vestmannaeyjum og Iceprotein á Sauðárkróki er nú að ljúka. Þar voru þróaðar nokkrar frumgerðir af vörum úr íslensku sjávarfangi og bætt í þær lífefnum eins og þörungaþykkni með skilgreinda lífvirkni, hýdrólysötum til að auka próteininnihald og lýsi til að auka ómega‐3 fitusýrur. Niðurstöðurnar sýna að vel er hægt að auka magn ómega‐3 fitusýra í fiskibollum án þess að það komi niður á bragðgæðum. Sama má segja um íblöndun þörungadufts og einnig tókst vel að auka próteinmagn í fiskbollunum.   Neytendakannanir voru framkvæmdar til að kanna smekk neytenda fyrir frumgerðum í samanburði við hefðbundna vöru sem þegar er á markaði.   Upplýsingar um lífvirku efnin og virkni þeirra hafði áhrif á hvernig fólki geðjaðist að vörunum. Áhrif upplýsinga voru háð ýmsum þáttum, eins og viðhorfum til heilsu og matar og viðhorfum til innihaldsefna í þeirri vöru sem prófuð var. Neytendakönnun á netinu sem yfir 500 manns tóku þátt í sýndu að fólk er almennt jákvæðara gagnvart auðgun ef um er að ræða þekkt hollustuefni á borð við ómega‐3. Einnig að betra er að veita upplýsingar um virkni þó að um þekkt efni sé að ræða, þar sem það eykur á jákvæða upplifun fólks af vörunni. Auðgun með þara virðist einnig vera raunhæfur kostur þar sem upplýsingar um notkunargildi þarans í vöru voru gefnar og svipað má segja varðandi fiskiprótein. Þessar vörur höfða almennt frekar til fólks sem leggur áherslu á hollustu matvæla, sem er nokkuð stór hópur samkvæmt þessum niðurstöðum. Almennt má álykta út frá þessum niðurstöðum að auðgun sjávarrétta sé raunhæfur möguleiki en huga þarf að merkingum og upplýsingagjöf til neytenda. Prototypes of seafood dishes enriched with bioactive compounds from the ocean, such as seaweed, fish proteins and fish oil to increase omega‐ 3 fatty acids have been developed to meet market demand. The results show that it is possible to increase the content of omega‐3 fatty acids in fishcakes without negatively affecting the flavour. Also the enrichment of seaweed and fish proteins to increase protein content was successful. Consumers were asked about liking of various prototypes compared to traditional fish dishes. Information on the added compounds and their bio‐activity affected the liking of the consumers. Influence of information depended on various factors like attitudes towards health, food and the added ingredients.   Web‐based consumer survey (500 respondents) showed that consumers were more positive towards enrichment of seafood if well‐known ingredients like omega‐3 were used. The information on health‐effect and bio‐activity was also positive regarding the consumer experience. even though the ingredients was well‐known. Enrichment using seaweed or proteins also seems to be a realistic option based on information of the health effect given. These kinds of products appeal more to consumers emphasising health benefits of their food. It can be stated from the results of the project that enrichment of seafood is a realistic option but labelling and information to consumers is important.

Markmið verkefnisins var að leita leiða til þess að bæta lifun og gæði lirfa þorsks og lúðu og nota til þess umhverfisvænar aðferðir. Markmiðið var einnig að opna fyrir möguleika á nýtingu ufsapeptíða sem aukið gæti verðmæti ufsa. Niðurstöður fyrra verkefnis í lúðueldi voru lofandi og bentu til þess að hentugast væri að meðhöndla lirfur með peptíðum í gegnum fóðurdýr auk þess sem nauðsynlegt væri að rannsaka frekar styrk meðhöndlunar. Í tengslum við verkefnið hefur verið þróuð og sett upp ný aðstaða í fóðurdýraræktun hjá Fiskey hf. til rannsókna á mismunandi meðhöndlunum fóðurdýra og stuðla þannig að auknum stöðugleika í framleiðslu lúðuseiða. Framkvæmdar hafa verið endurteknar tilraunir með lífvirkum efnum í ræktun hjóldýra og virtust þau þola vel ákveðinn styrk efnanna. Helstu niðurstöður tilrauna á fyrstu stigum þorskeldis benda til þess að meðhöndlun með ufsapeptíðum skili sér í góðum vexti, áberandi hraðari þroska innri líffæra og mun lægri tíðni galla í lirfum. Þó er ljóst að rannsaka þarf frekar áhrif mismunandi styrks meðhöndlunar. Sterkar vísbendingar eru um að IgM og lysozyme séu til staðar í þorsklirfum fljótlega eftir klak eða mun fyrr en áður hefur verið haldið fram auk þess sem meðhöndlun virtist örva framleiðslu þeirra. Meðhöndlun með ufsapeptíðum virðist ekki hafa áhrif á samsetningu bakteríuflóru lirfa en ákveðin tegundasamsetning greindist í meltingarvegi lirfa í kerjum þar sem lifun og gæði lirfa voru best. Gefur þetta vísbendingar um að ákveðin tegundasamsetning bakteríuflóru sé þorsklirfum hagstæð.

The main goal of this project was to increase viability and quality of cod and halibut larvae before and during the first feeding period by using bioactive products. The aim was also to increase the exploitation and value of pollock. The findings of previous projects in halibut culture were promising and indicated that treating live feed is a suitable method to carry bioactive products to the larval intestines during first feeding but the intensities of treatment needed to be further investigated. New facilities have been developed in relation to the project for research in the live feed culture at Fiskey Ltd. to promote increased stability in the production of halibut fingerlings. Repeated experiments have been conducted in the culture of rotifers and results indicates good tolerance towards treatment with bioactive products in certain intensities. The overall results of the project indicated that pollock peptides may promote increased growth and quality of cod larvae during first feeding. The results also indicate the presence of IgM and lysozyme early post hatching, but it has not been observed in cod larvae of this size before. Furthermore, results also indicate that hydrolysates from pollock can stimulate the production of these factors in cod larvae. Treatment using pollock peptides, did not affect the bacterial community structure of live feed or cod larvae, however a similar structure was observed in larvae from the most successful production units different from other tanks. The results therefore indicate a bacterial community structure that may be preferable to the cod larvae.

Notkun lífvirkra efna í lúðueldi

Megin markmið verkefnisins var að stuðla að aukinni afkomu lúðulirfa í eldi og nota til þess umhverfisvænar aðferðir. Notuð voru lífvirk efni sem auðvelt var að nálgast, stuðluðu að auknu verðmæti sjávarfangs og hefðu jafnframt einhverja þá virkni sem óskað var eftir þ.e. bakteríudrepandi/-hamlandi, prebiotik eða ónæmisörvandi virkni. Gerðar voru tilraunir með ýmis efni í verkefninu þ.e. kítósan afleiður auk peptíða sem unnin voru úr kolmunna, þorski og ufsa. Áhrif meðhöndlunar með efnunum voru metin m.t.t. vaxtar og afkomu lirfa og fóðurdýra svo og m.t.t. samsetningar bakteríuflóru og örvunar ósérhæfðrar ónæmissvörunar í lirfum. Helstu niðurstöður benda til að hentugasta aðferðin til að koma efnum í lirfur er að nota fóðurdýr (Artemia) og var í verkefninu þróuð aðferð til að meðhöndla þau. Lífvirku efnin virtust ekki hafa bakteríuhamlandi áhrif í eldisumhverfi fóðurdýranna en stuðla að breyttri samsetningu bakteríuflórunnar. Lífvirk efni virtust fyrst og fremst nýtast sem bætiefni þar sem fóðurdýr voru bústin og spræk. Afkoma og gæði lirfa í eldiseiningum Fiskey hf. er mjög mismunandi og eru engin augljós tengsl á milli afkomu á kviðpokastigi og afkomu og gæða lirfa í lok startfóðrunar. Samsetning bakteríuflóru reyndist einnig mjög mismunandi í kviðpokalirfum og lirfum í startfóðrun. Framkvæmdar voru þrjár aðskildar tilraunir í seiðaeldisstöð Fiskeyjar þar sem lirfur í startfóðrun voru meðhöndlaðar með lífvirkum efnum. Helstu niðurstöður leiddu í ljós að mikilvægt er að meðhöndla með réttum styrk efna og í hæfilega langan tíma þar sem of mikill styrkur getur haft neikvæð áhrif á vöxt og myndbreytingu lirfa. Meðhöndlun með kolmunnapeptíðum þótti gefa lofandi niðurstöður og hafa góð áhrif á myndbreytingu lirfa. Lífvirk efni virtust ekki hafa afgerandi áhrif á fjölda ræktanlegra baktería í meltingarvegi lirfa en meðhöndlun með kolmunna- og þorskpeptíðum gæti hugsanlega breytt samsetningu flórunnar. Rannsóknir á ósérhæfðri ónæmissvörun lúðulirfa leiddu í ljós tilvist C3 og Lysozyme frá lokum kviðpokastigs en framleiðsla á IgM hefst ekki fyrr en um 28 dögum eftir að startfóðrun hefst. Meira magn IgM mældist á fyrstu vikunum í lirfum sem meðhöndlaðar voru með ufsapeptíðum og gæti það bent til ónæmisörvandi áhrifa. Niðustöður verkefnisins í heild sinni benda til þess að þau lífvirku efni sem rannsökuð voru hafi ekki afgerandi áhrif á bakteríuflóru eldisins en meðhöndlun lirfa í startfóðrun með réttum styrk lífvirkra efna gæti haft góð áhrif á afkomu og vöxt lirfa og örvað ósérhæfða ónæmissvörun lirfa á þessu viðkvæma stigi eldisins þegar þær hafa enn ekki þróað sérhæft ónæmissvar.

The aim of this project was to promote increased survival of halibut larvae during first feeding by using bioactive products. The bioactive products were selected by the criterion that they were easily accessible and induced any of the desired effects i.e. inhibiting bacterial growth, prebiotic effects or immunostimulants. The products studied are chitosan and peptide hydrolysates from blue whiting, cod and saithe. The effects of treatment were evaluated with respect to growth and survival of larvae and the live feed (Artemia) as well as effects on bacterial numbers or the community structure of the intestinal microbiota of larvae and stimulation of the innate immune system of the larvae. The results indicate that treating live feed (Artemia) is a suitable method to carry the bioactive products to the larval intestines during first feeding and a new technique has been standardized for treatment of the live feed with the products. The bioactive products did not affect the total bacterial count in the Artemia but the composition of the bacterial community may be changed as a result of the treatment. The Artemia seems to use the bioactive products as a food supplement and was well suited to be used as live feed. A significant variation in overall success of larvae was observed without any obvious correlation between survival of larvae at the end of the yolk sac stage and at the end of first feeding. A different bacterial pattern was observed in the intestine at the yolk sac stage compared to first feeding larvae. Three separate experiments were carried out in the halibut production units at Fiskey Ltd. where larvae were treated with various bioactive products. The results emphasize the importance of treating larvae with the appropriate concentrations of the products, as elevated concentrations can negatively affect growth and metamorphosis of the larvae. Treatment with peptides from blue whiting resulted in relatively good survival of larvae with similar success of metamorphosis compared to control units. The bioactive products did not affect bacterial growth but there were indications that peptides from blue whiting and cod may affect the composition of the intestinal community of bacteria in the larvae. Results from studies of the immunological parameters indicate the presence of C3 and Lysozyme already from the end of the yolk sac stage and the initialization of IgM production after approximately 28 days in feeding. Production of IgM was stimulated in larvae treated with peptides from saithe, indicating immunostimulating effects of this product. The overall results indicate that the bioactive products studied did not affect the bacterial flora during the first production stages of halibut larvae. However, if used in the appropriate quantities and at the right time, the products may promote survival and growth and stimulate the innate immunity of larvae.
